MOTS-c — The Mitochondrial Peptide
What Is It?
MOTS-c is a 16-amino acid peptide encoded within the mitochondrial genome — specifically within the 12S ribosomal RNA gene. Discovered in 2015 by researchers at the University of Southern California led by Dr. Pinchas Cohen, it represents a novel class of signalling molecules known as mitochondrial-derived peptides (MDPs).
In plain terms: it’s a signal your own mitochondria — the energy-producing engines of your cells — send out to regulate metabolism across the body. And like many of the body’s natural repair signals, MOTS-c levels in plasma decline significantly with age — being 11% higher in young people compared to middle-aged, and 21% higher compared to older individuals.

What Are Researchers Studying It For?
🏃 Exercise Mimetic & Physical Performance One of the most exciting areas of MOTS-c research. In humans, exercise induces endogenous MOTS-c expression in skeletal muscle and in circulation — suggesting it may be one of the key signals responsible for exercise’s metabolic benefits.
🩸 Insulin Sensitivity & Diabetes MOTS-c targets skeletal muscle and can enhance glucose metabolism, playing an important role in the regulation of diabetes and insulin resistance. Research in humans has found that circulating MOTS-c levels are lower in type 2 diabetes patients compared with healthy controls
❤️ Cardiovascular Health Recent studies have shown a protective effect of MOTS-c against cardiac dysfunction and pathological remodelling, with research demonstrating it can prevent the development of heart failure through activation of the AMPK pathway. A 2025 study also found it restores mitochondrial respiration in the diabetic heart.
🧬 Longevity & Healthy Ageing MOTS-c has shown beneficial effects on age-related diseases including diabetes, cardiovascular disease, osteoporosis, and Alzheimer’s — with research suggesting that the age-related decline in physical capability may potentially be reversed with MOTS-c treatment.
